چکیده
The rlrA pathogenicity islet in Streptococcus pneumoniae TIGR4 encodes three surface proteins, RrgA, RrgB, and RrgC, and three sortase enzymes. Using transmission electron microscopy, cell fractionation, cell wall sorting signal domain swapping, and Western blotting, we show that RrgA and RrgB are incorporated into a multisubunit pilus in S. pneumoniae.

Roles of the sortases of Streptococcus pneumoniae in assembly of the RlrA pilus.
Pili have been observed on the surface of several gram-positive bacteria, including Streptococcus pneumoniae. The S. pneumoniae strain TIGR4 pilus is composed of three structural subunit proteins encoded in the rlrA pathogenicity islet, RrgA, RrgB, and RrgC. RrgB comprises the pilus backbone, RrgA is observed at intervals along surface pili, while RrgC is found in a loosely defined relationship...

The rlrA genetic islet encodes an extracellular pilus in the Gram-positive pathogen Streptococcus pneumoniae. Of the three genes for structural subunits, rrgB encodes the major pilin, while rrgA and rrgC encode ancillary pilin subunits decorating the pilus shaft and tip. Deletion of all three pilus-associated sortase genes, srtB, srtC and srtD, completely prevents pilus biogenesis. Expression o...
متن کاملPilus Operon Evolution in Streptococcus pneumoniae Is Driven by Positive Selection and Recombination
BACKGROUND The evolution of bacterial organelles involved in host-pathogen interactions is subject to intense and competing selective pressures due to the need to maintain function while escaping the host immune response. To characterize the interplay of these forces in an important pathogen, we sequenced the rlrA islet, a chromosomal region encoding for a pilus-like structure involved in adher...
متن کاملTranscriptional regulation in the Streptococcus pneumoniae rlrA pathogenicity islet by RlrA.
The proper temporal expression of virulence genes during infection is crucial to the infectious life cycle of microbial pathogens, particularly in pathogens that encounter a multitude of environments in eukaryotic hosts. Streptococcus pneumoniae normally colonizes the nasopharynges of healthy adults but can cause a range of diseases at a variety of host sites. Transcriptional regulators that ar...
متن کاملMolecular architecture of Streptococcus pneumoniae TIGR4 pili.
Although the pili of Gram-positive bacteria are putative virulence factors, little is known about their structure. Here we describe the molecular architecture of pilus-1 of Streptococcus pneumoniae, which is a major cause of morbidity and mortality worldwide. One major (RrgB) and two minor components (RrgA and RrgC) assemble into the pilus. Results from TEM and scanning transmission EM show tha...
